Insight into the Substrate Specificity of Lactobacillus paracasei Aspartate Ammonia-Lyase

Aspartate ammonia-lyase (AAL) catalyzes the reversible conversion reactions of aspartate to fumaric acid and ammonia. In this work, Lactobacillus paracasei LpAAL gene was heterologously expressed in Escherichia coli. As well as a recombinant His-tagged protein, maltose-binding protein (MBP) fused used enhance its solubility expression level. Both proteins showed broad substrate specificity, catalyzing aspartic acid, phenylalanine, tyrosine produce trans-cinnamic p-coumaric respectively. The optimum reaction pH temperature for four substrates were measured at 8.0 40 °C, Km values 5.7, 8.5, 4.4, 1.2 mM, kcat 6.7, 0.45, 4.96, 0.02 s−1, Therefore, are bona fide enzyme.
